彈奏前要先調音，使用電子調音器是最簡單的方法。把調音器夾在琴頭上，然後輕輕的撥弦，調音器便會顯示當下的音高，這時可轉動琴頭的琴鈕調音了，把弦線收緊就會把音升高而放鬆弦線就能把音降低。按下圖調音，傳統G弦不是最低音，弦線是比較幼，C弦是最低音最粗。記緊G弦不是最低音，這是傳統Ukulele的特色。如果G弦是最粗或是鋼弦，這種弦就要調Low G(低一個8度)。

The standard tuning for soprano, concert, and tenor ukuleles is C-tuning, g, c, e, a. The g string is tuned an octave higher than might be expected. This is known as re-entrant tuning. Some prefer "Low G" tuning, with the G in sequence an octave lower
